饮用水中的真菌污染会引起水体产生嗅味,其潜在致病性容易导致流行性疾病的发生.以西北某市水源井水为研究对象,利用PCR-DGGE分子指纹图谱技术分析水体中的真菌多样性.研究得出,该地区地下水中真菌种类较为丰富,真菌多样性指数、丰富度指数和均匀度指数分别为0.89~1.33、9~25 和0.405~0.460. 不同样点真菌群落相似性存在一定关系,相邻样点间相似性较高.对DGGE条带进行回收、克隆、测序,显示隐球菌属(Cryptococcus.sp)、暗球腔菌属(Pluteus.sp)、镰刀菌属(Fusarium.sp)、青霉属(Penicillium.sp)四种类型为优势种属.相关性分析表明不同种属的真菌与环境变量的相关性并不相同,pH值和温度和对镰刀菌属(Fusarium.sp)、青霉属(Penicillium.sp)的生长相关性较大. 研究为地下水真菌污染控制提供了一定的理论依据.
The occurrence of fungi in drinking-water may be linked to taste, odour and some health problems, and its potential pathogenicity can easily lead to the occurrence of epidemic diseases. Water samples were taken from twelve ground water source wells, then PCR-DGGE molecular fingerprinting technology was used to analyze the fungal diversity. The results showed that there were abundant fungal species in groundwater in this area, with fungal diversity index, richness index and evenness index ranged from 0.89~1.33, 9~25 and 0.405~0.460, respectively. There was a certain relationship between the similarity of fungal communities in different sites, and the similarity between adjacent spots was higher. The DGGE band was recovered, cloned, and sequenced, and the results showed that Cryptococcus sp., Pluteus sp., Fusarium sp., Penicillium sp. Species types were dominant species. Correlation analysis showed that the correlation between different species of fungi and environmental variables was not the same, pH and temperature are more relevant to the growth of Fusarium sp. and Penicillium.sp. The research provides a certain theoretical basis for the control of groundwater fungal pollution.
